The Lauren&#8217;s classification distinguishes between a well differentiated intestinal\u00a0type and a poorly differentiated diffuse type of the\u00a0disease, which differ in their development and the\u00a0characteristic sequence of genetic changes. The\u00a0purpose of this first study done on a selected\u00a0sample of Slovene patients was to investigate the\u00a0allele loss of several tumor suppressor genes\u00a0(<\/span><em style=\"font-size: 13px;line-height: 19px\">APC, p53, Rb, nm23 and DCC<\/em><span style=\"font-size: 13px;line-height: 19px\">) and thus distinguish between the intestinal and diffuse type\u00a0of gastric adenocarcinoma in the Slovene population. Another aim of this work was to introduce\u00a0the newest method for investigating the allele loss.\u00a0Seventeen randomly selected frozen samples of\u00a0adenocarcinomas and adjacent normal tissues\u00a0were obtained from 17 patients treated by surgery\u00a0in 1997. The polymerase chain reaction was\u00a0used for multiplying microsatellite sequences of\u00a0the tumor suppressor genes. The obtained PCR\u00a0products were analysed with the capillary electrophoresis in the automatic gene analyser. Loss\u00a0of heterozigosity was found in 17 % of samples\u00a0for <\/span><em style=\"font-size: 13px;line-height: 19px\">APC<\/em><span style=\"font-size: 13px;line-height: 19px\">, in 50 % for<\/span><em style=\"font-size: 13px;line-height: 19px\"> p53<\/em><span style=\"font-size: 13px;line-height: 19px\">, and in 100 % for <\/span><em style=\"font-size: 13px;line-height: 19px\">DCC<\/em><span style=\"font-size: 13px;line-height: 19px\">.\u00a0There was no loss of heterozigosity for <\/span><em style=\"font-size: 13px;line-height: 19px\">Rb<\/em><span style=\"font-size: 13px;line-height: 19px\"> and\u00a0<\/span><em style=\"font-size: 13px;line-height: 19px\">nm23<\/em><span style=\"font-size: 13px;line-height: 19px\">. Dfferences in genetic alterations between\u00a0the intestinal and diffuse type of adenocarcinoma in the Slovene population were identified. The\u00a0results represent a good starting point for the multiplex polymerase chain reaction using a larger\u00a0set of genetic markers, and for its application in\u00a0the research of some other, not yet investigated\u00a0tumor suppressor and mismatch repair genes.<\/span><\/p>\n","protected":false},"excerpt":{"rendered":"<p>Stomach cancer (gastric adenocacinoma) is a common malignant disease in\u00a0Slovenia.
