Determination of estrogen receptors in small intestinal mucosa

Small intestinal mucosa of rats was tested for the presence of estrogen receptors. For determination of estrogen receptors  3H-estradiol 17-beta binding assay with or with our excess 1H-estradiol 17-beta was used. Three sub cellular fractions including nuclei, microsomes and cytosol, were analyzed. Some animals were pretreated with 1H-estradiol in order to determine whether it induces an increase in specific binding of estradiol. Estrogen receptors were never found, either in cytosol or in microsomes. The nuclear fraction obtained from the intestinal mucosa of non.treated animals, showed low specific binding (63fmol/mg of protein), while in rats treated with estradiol this value increased by approx. nine times as compared to control values (600fmol/mg).
